The draw for the event was conducted at 11am local time on Thusday, 4 August. Serena Williams was the defending champion, but she lost in the third round to Elina Svitolina. Mónica Puig won Puerto Rico's first ever Olympic gold medal, defeating Angelique Kerber in the final, 6–4, 4–6, 6–1.
